Uracil-DNA Glycosylase (UNG or UDG) is derived from recombinant cloning and expression of Escherichia coli, with a molecular weight of 25 kDa. It can catalyze the release of free uracil in single-stranded and double-stranded DNA containing uracil, and has no activity on RNA. It can be used to prevent contamination caused by PCR amplification products. Its principle is based on: if dUTP is substituted for dTTP in PCR reaction to form PCR amplification products containing dU bases, this enzyme can selectively break the glycosidic bond of U bases in single-stranded and double-stranded DNA, degrading the PCR amplification product.
