Capping Efficiency is an important quality attribute indicator for various types of mRNA drugs, including mRNA vaccines, and is a required validation document for the registration of mRNA products. Currently, the measurement of capping efficiency is mainly conducted through liquid chromatography-mass spectrometry (LC-MS). There is a difference of hundreds in molecular weight between capped mRNA and uncapped mRNA, which makes it difficult to precisely analyze the difference between capped and uncapped samples compared to the molecular weight of complete mRNA, which is in the hundreds of thousands. Therefore, to analyze capping efficiency, it is first necessary to process the long mRNA strands into smaller fragments, turning the challenge of finding a difference of hundreds within a molecular weight of hundreds of thousands into a difference of hundreds within a molecular weight of a few thousand.
