Deoxyribonuclease I (DNase I) is a nonspecific endonuclease that can digest single-stranded or double-stranded DNA. It hydrolyzes phosphodiester bonds, producing single deoxyribonucleotides and oligodeoxyribonucleotides containing a 5'-phosphate group and a 3'-OH group. The activity of DNase I depends on Ca2+ and can be activated by divalent metal ions such as Mn2+, Mg2+, Zn2+, etc. In the presence of Mg2+, DNase I randomly cleaves double-stranded DNA at any site; in the presence of Mn2+, DNase I can cut DNA double strands at the same site, forming blunt ends or sticky ends with 1-2 nucleotides protruding.
This kit is based on the nonspecific cleavage of single-stranded or double-stranded DNA by DNase I. It is used in conjunction with Biori's nucleic acid extraction or purification kits to establish a rapid preparation of RNA samples without DNA, which can be used for subsequent detection of total RNA in bioproducts. RNA samples can be prepared in as fast as 1 hour.
