DNase I (Deoxyribonuclease I) is an enzyme that efficiently digests single- or double-stranded DNA fragments.The activity of DNase I is very dependent on the Ca2+ level and can be activated by either Mg2+ or Mn2+. In the presence of Mg2+, DNase I randomly shears DNA fragments from any site; in the presence of Mn2+, DNase I shears double stranded DNA at approximately the same site, resulting in either a flat-ended DNA fragment or a sticky end with 1-2 nucleotides protruding from it. DNase I has optimal endonuclease activity in the pH range of 7-8 and can be used to remove single- or double-stranded DNA fragments from a variety of RNA samples.
