Restriction Endonucleases

Fast-cut restriction endonucleases that complete site-specific DNA digestion within 5-15 min and retain 100% activity in the universal 10× Cut-Buffer.

Restriction Endonucleases
Selection guide

Pick the enzyme by end type and site

All fast-cut enzymes retain 100% activity in the universal 10× Cut-Buffer, so any two can be combined without buffer exchange. Each row names a first pick for one type of end or site and lists the other enzymes of the same type; the full recognition-site table follows the product list.

Frequently asked questions

Can two of these enzymes be used in the same reaction?
Yes. All fast-cut enzymes retain 100% activity in the universal 10× Cut-Buffer, so any two work together without buffer exchange. Keep the combined enzyme volume below one-tenth of the reaction volume. If the two enzymes have different optimal temperatures, digest with the lower-temperature enzyme first, then add the second enzyme and continue at the higher temperature.
How long does a digestion take?
Incubate for 15 min (plasmid DNA), 15-30 min (PCR products) or 30-60 min (genomic DNA). Most enzymes work at 37°C; TaqI is used at 65°C and PspGI at 75°C. Digestion for longer than 3 h is not recommended, as prolonged incubation may lead to star activity.
Which enzymes cannot be heat-inactivated?
AvrII, BamHI, BglII, FspI, KpnI, MunI, NaeI, NruI, PspGI, PstI, PvuII, RsaI, TaqI. Purify the digest by phenol-chloroform extraction or column purification instead. The other 47 enzymes are inactivated at 80°C for 20 min.
Which products are in the Restriction Endonucleases category?
