AcuGenix™ Magnetic Sputum Nucleic Acid Extraction Kit
Specialized Pathogen Nucleic Acid Co-Isolation for High-Mucin Respiratory Samples | Superior Sensitivity for Refractory TB, Influenza & RSV

Product overview
The AcuGenix™ Magnetic Sputum Nucleic Acid Extraction Kit is a magnetic bead-based co-isolation solution engineered exclusively for pathogen nucleic acid extraction from respiratory sputum. Optimized to address the inherent challenges of sputum—including high mucin content, elevated host cellular background, and complex PCR inhibitor profiles—it employs proprietary silica-functionalized superparamagnetic beads and a pre-optimized buffer system to efficiently lyse mucin matrices, digest host cells, and rapidly release pathogen-derived DNA and RNA.
Features and applications
- Sputum-Optimized Workflow: Purpose-built for viscous, inhibitor-rich sputum samples, eliminating the need for manual mucin digestion or additional pre-treatment steps common with generic extraction kits.
- Proprietary Bead-Buffer System: Silica-functionalized superparamagnetic beads deliver high nucleic acid binding affinity under high-salt/low-pH conditions; simple salt adjustment enables efficient elution without phenol-chloroform extraction, ethanol precipitation, or spin-column handling.
- Closed-Tube Biosafety Design: Entire workflow is performed in a closed system, minimizing aerosol contamination risks and meeting BSL-2 laboratory compliance requirements.
- Ultra-High Sensitivity for Trace Pathogens: Maintains consistent recovery efficiency for target pathogens down to 10 copies per reaction, including refractory-to-lyse Mycobacterium tuberculosis, Influenza virus, and respiratory syncytial virus (RSV).
- Exceptional Purity for Reliable Downstream Results: Rigorous batch validation confirms stable yield and high purity, with efficient removal of mucins, host genomic DNA, and respiratory secretion-derived PCR inhibitors that cause false-negative results.
- Seamless Ecosystem Compatibility: Purified nucleic acids require no additional cleanup and integrate fully with the AcuGenix™ portfolio of reverse transcription and NGS library preparation reagents.
- Unbiased pathogen detection in lower respiratory tract infections via metagenomic next-generation sequencing (mNGS)
- Multiplex respiratory pathogen panel (RPP) testing for clinical research
- Drug susceptibility genotyping for Mycobacterium tuberculosis and other refractory pathogens
- Influenza, RSV, and other respiratory virus surveillance studies
- Host-pathogen interaction research using sputum-derived nucleic acids
- Method development for clinical molecular diagnostics of respiratory infections

Performance data
Broad Pathogen Compatibility

Each grade corresponds to a defined sputum viscosity category. Following spiking of 10 copies of Mycobacterium tuberculosis genomic standard into sputum samples across all grades, BR2D103 reliably detected the 10-copy target in every sample type.
Note: BR2D103 is the catalog number for this product. Mycobacterium tuberculosis is a refractory target in clinical respiratory pathogen testing; these results confirm compatibility with the full spectrum of clinical sputum samples, from low- to high-viscosity.
Rapid Sputum Liquefaction

BR2D103 maintained stable detection of the 10-copy target across two high-viscosity sputum grades, with liquefaction durations ranging from 5 min to 20 min, accommodating variable workflow timelines.
Note: Laboratory throughput varies widely; tolerance to extended liquefaction times prevents assay failure due to processing delays, improving workflow robustness.
Excellent Inter-Batch Consistency

Three independent reagent lots of BR2D103 were used to extract nucleic acids from three distinct clinical sputum samples. Ct values for the same sample were highly consistent across all lots, confirming excellent batch-to-batch reproducibility.
Note: Lot-to-lot consistency is a critical quality metric for clinical diagnostic reagents, ensuring reliable and comparable results across multiple batches and multicenter studies.

Ten copies of pathogen template were spiked into sputum samples of varying grades. For challenging sample types including Level IV purulent sputum and Level V bloody sputum, BR2D103 delivered overall superior target detection Ct values compared to leading competitor kits, demonstrating enhanced analytical sensitivity and inhibitor tolerance.
Note: Purulent and bloody sputa contain high levels of mucoproteins and heme components that inhibit PCR; these results confirm BR2D103 effectively overcomes interference from complex clinical matrices, improving diagnostic sensitivity.
