EcoR I GMP-grade, GMP-BP-E16, is a GMP-grade restriction endonuclease for DNA digestion workflows in mRNA and IVT process development. Its Type II cleavage mechanism produces 5′ cohesive overhangs. It recognizes the EcoR I site and supports plasmid linearization or preparation of DNA fragments with defined sticky ends.

Specifically recognizes the palindromic sequence GAATTC and cleaves at the G/A site, producing 5′ cohesive overhangs.
GMP-grade EcoR I restriction endonuclease.

1 Digestion of plasmid by varying units of EcoR I GMP-grade

Description: To compare the digestion performance of EcoR I GMP-grade from Biori and Supplier A on 1 μg of plasmid containing a single EcoR I recognition site, using varying enzyme amounts.

Conclusion: EcoR I GMP-grade supplied by Biori demonstrated high activity and superior digestion performance compared with Supplier A, particularly at low enzyme concentrations.

2 EcoR I Lot-to-Lot and Accelerated Stability

Description: To evaluate the lot-to-lot stability (three lots) and accelerated stability (stored at 25 °C for 32 days) of Biori EcoR I GMP-grade. The activity of three lots of Biori EcoR I GMP-grade was measured after storage at 25 °C for 32 days.

Conclusion: After 32 days at 25 °C, the activity of all three lots of Biori EcoR I GMP-grade was nearly identical to that at Day 0, with no significant decrease and little lot-to-lot variation.
