BspQ I GMP-grade, GMP-BP-E09, is a GMP-grade type IIS restriction endonuclease. Its Type IIS cleavage mechanism can generate a “scarless” template, ensuring that the IVT product contains no extra nucleotides. It recognizes the BspQ I site and supports plasmid linearization or preparation of DNA fragments with defined sticky ends.

The cleavage site is located outside the recognition sequence, generating a 5′ overhang.

1 Plasmid Adaptability and lot-to-lot Stability

Description: Three different plasmids, each containing a single BspQ I GMP-grade recognition site, were tested. Under identical reaction conditions and incubation time, the cleavage activity of three lots of Biori BspQ I GMP-grade was compared with that of Supplier N.

Conclusion: Multiple lots of BspQ I GMP-grade demonstrated consistent cleavage performance across different plasmids, confirming good lot-to-lot stability and broad adaptability to various plasmid templates.

2 4°C storage stability

Description: The activity of Biori BspQ I GMP-grade was measured at 7, 14, 21, and 28 days of storage at 4°C.

Conclusion: Biori BspQ I GMP-grade retained most of its activity throughout the 28-day storage period at 4°C, with only a slight decrease relative to the -20°C sample.

3 BspQ I linearization: downstream IVT comparison (Biori vs Supplier N)

Description: To compare the performance of linearized templates prepared with Biori BspQ I GMP-grade versus Supplier N in downstream in vitro transcription (IVT) applications. Gel electrophoresis analysis of IVT products generated from linearized templates prepared with Biori BspQ I GMP-grade or Supplier N.

Conclusion: Linearized templates prepared with Biori BspQ I GMP-grade, when used with Biori's T7 high-efficiency transcription kit (Cat: BP-01-50), yielded IVT products with high integrity, comparable to those obtained with Supplier N.
