RNase R GMP-grade
RNase R enzyme for digesting linear RNA.

Product overview
RNase R (Ribonuclease R) is a magnesium-dependent ribonuclease derived from the E. coli RNR superfamily, which completely hydrolyzes linear RNA in the 3'→5' direction into dinucleotides and trinucleotides. It is insensitive to circular RNA and double-stranded RNA, making it suitable for the production of specialized RNA structures such as circular RNA (circRNA), lariat RNA, double-stranded RNA with fewer than 7 nucleotides at the 3' protruding end, and structurally complex tRNAs. RNase R is widely used in gene expression and alternative splicing studies to digest linear RNA, thereby enriching circRNA or lariat RNA.

Performance data
1 Functional test
Description: To compare the preRNA-digestion specificity of Biori RNase R and Supplier K, equivalent amounts of preRNA and circRNA were incubated with equal enzyme units for the same duration. The ability of each enzyme to digest preRNA while leaving circRNA intact was then assessed.

Conclusion: When the enzyme amount was increased to 10 U, the majority of circRNA remained intact after digestion with Biori RNase R, whereas Supplier K RNase R caused noticeable degradation of circRNA.
2 Stability Assay
Description: The digestion performance of Biori RNase R (stored at 4 °C) was compared with that of the control (stored at -20 °C). Equal amounts of preRNA and circRNA were incubated with equivalent enzyme units for the same duration, and the extent of preRNA digestion versus circRNA resistance was assessed by gel electrophoresis.

Conclusion: Biori RNase R retained full activity after 1-month storage at 4 °C, showing no detectable difference in its ability to digest preRNA compared with the -20 °C control.
3 Lot-to-lot consistency of RNase R activity
Description: Equivalent amounts of preRNA and circRNA were incubated with equal enzyme units from each of the five lots for the same digestion time. The ability of each lot to digest preRNA while preserving circRNA was assessed by gel electrophoresis.

Conclusion: All five production lots of Biori RNase R demonstrated consistent lot-to-lot stability and activity, as evidenced by efficient digestion of preRNA and effective enrichment of circRNA.
