RNase Residual Quantitative Detection Kit, BP-06
Quantifies residual RNase activity in mRNA workflows.

Product overview
Ribonuclease (RNase) is a class of nucleases that catalyzes RNA degradation into small fragments. RNase A is a widely used endonuclease that specifically cleaves phosphodiester bonds on single-stranded RNA at the 3' end of cytidine (C) and uridine (U) residues, producing oligonucleotides with 2',3'-cyclic phosphate termini. Biori's RNase Residual Quantitative Detection Kit employs a fluorescent probe-based approach for rapid, high-sensitivity detection of RNase A activity. The kit utilizes a synthetic RNA oligonucleotide probe with a FAM fluorophore (donor) at one end and a TAMRA quencher (acceptor) at the other. When the two groups are in close proximity, fluorescence energy transfers from donor to acceptor, quenching the donor's fluorescence. Upon cleavage by RNase, the probe separates, the two groups disperse, and FAM fluorescence is no longer quenched, resulting in detectable fluorescence. The rate of fluorescence signal increase correlates positively with enzyme quantity and activity.

Performance data
1 Assay principle
Description: Schematic diagram of the Biori RNase Residual Quantitative Detection Kit.

Conclusion: The rate of fluorescence signal increase correlates positively with enzyme quantity and activity.
2 Sensitivity
Description: The sensitivity of the kit was evaluated by detecting varying amounts of RNase A.

Conclusion: The kit exhibits a sensitivity of 0.078 pg RNase.
3 Fluorescence intensity plot
Description: Time course of fluorescence intensity for the detection of various amounts of RNase A using this kit over a 60-minute period.

Conclusion: The figure shows the changes in fluorescence intensity of RNase A over a 60-minute period.
4 Standard curve
Description: Establishing the standard curve by gradient dilution of RNase A standard.

Conclusion: The RNase A standard were subjected to gradient dilution for fluorescence measurement. The data were processed using linear fitting. The results showed that the linear R2 value was greater than 0.99.
