Product overview

AcuGenix™ AmpSeq Library Amplification Mix is a 2× ready-to-use master mix optimized for NGS library amplification workflows. All core reaction components—including a directed evolution-derived AmpHifi HS high-fidelity hot-start DNA polymerase, optimally formulated dNTPs, proprietary reaction buffer, and amplification enhancers—are pre-mixed for immediate use, eliminating the need for additional reagent supplementation and minimizing pipetting errors.

Features and applications

Features
  • Engineered High-Fidelity Performance: The AmpHifi HS polymerase, developed via directed evolution, delivers superior broad-spectrum template compatibility, an ultra-low base substitution rate, and exceptional amplification sensitivity compared to wild-type enzymes. It efficiently amplifies challenging templates including GC-rich sequences, secondary structure-laden regions, and FFPE-derived nucleic acids, reducing amplification bias for complex samples.
  • Optimized Buffer System: The proprietary buffer incorporates background suppressors and amplification stabilizers to inhibit non-specific amplification and maintain reaction consistency, ensuring excellent run-to-run stability and amplicon abundance uniformity across all target regions.
  • Ready-to-Use Convenience: Pre-formulated 2× master mix streamlines workflow setup, reduces hands-on time, and minimizes batch-to-batch variability caused by manual reagent mixing.
  • Seamless Ecosystem Compatibility: Fully compatible with the entire AcuGenix™ portfolio of library preparation and multiplex PCR kits, enabling end-to-end workflow integration without protocol optimization.
Applications
  • Whole-Genome Sequencing (WGS) library amplification
  • Whole-Exon Sequencing (WES) library amplification
  • RNA Sequencing (RNA-seq) library amplification
  • Single-Cell Sequencing library amplification
  • Targeted Capture Sequencing library amplification
  • Amplicon-based NGS panel validation

Robust Yields Across a Wide Input Range

BR3M105 AcuGenix™ AmpSeq Library Amplification Mix: Figure 1. Superior amplification performance of BR3M105 across input levels
Figure 1. Superior amplification performance of BR3M105 across input levels. Libraries subjected to end repair were enriched using BR3M105 or commercial alternatives, with cycle numbers scaled to input (16/10/3 cycles). Following standardized bead cleanup, BR3M105 produced the highest library concentrations across all tested inputs. The yield advantage was most pronounced at 0.1 ng and 10 ng, where BR3M105 generated 30–40% more library than Supplier K and 2–3× more than Suppliers Y/V. These data confirm high amplification efficiency and reliable output from 0.1 ng to 1000 ng.

Note: BR3M105 is the catalog number for AcuGenix™ AmpSeq Library Amplification Mix. High-yield performance at low inputs is critical for library construction from single-cell, cfDNA, and other limited-sample workflows.

Figure 2. Sequencing performance and coverage uniformity of BR3M105. Libraries amplified with BR3M105 or commercial reagents were sequenced and analyzed. Coverage, depth, and base quality (Q30) were calculated using samtools; 1-kb genome binning (bedtools) yielded mean depth and Low Coverage Index (LCI). BR3M105 achieved parity with Supplier K on LCI while outperforming competitors in Q30 and sequencing depth. These results demonstrate that BR3M105 preserves library complexity and maximizes usable data output.

Note: Low Coverage Index (LCI) reflects uniformity of genomic coverage; lower LCI indicates reduced amplification bias. High Q30 scores indicate superior base-calling accuracy.

High Fidelity and Low Error Rate

Figure 3. Polymerase fidelity and error profiling of BR3M105. Libraries were sequenced and aligned with BWA. Overlapping read regions were parsed using samtools to evaluate bidirectional base-substitution rates. BR3M105 achieved fidelity parity with the Supplier K gold standard and showed reduced polymerase-induced errors compared to Suppliers V and Y. These data confirm superior amplification accuracy and strong potential as a premium domestic replacement for benchmark amplification modules.

Note: High fidelity is a critical metric for amplification reagents, directly impacting the reliability of variant detection. Low error rates are essential for sensitive detection of low-frequency mutations.
