Product overview

AcuGenix™ mRNA Capture Beads are superparamagnetic microbeads functionalized with surface-immobilized Oligo(dT), engineered for high-efficiency, high-purity isolation of intact poly(A)+ RNA from eukaryotic samples. Through a proprietary manufacturing process, the beads feature a tailored Oligo(dT) grafting density optimized for broad-spectrum mRNA capture, a uniform average particle size of ~1 μm, and stable dispersion in aqueous buffer systems with no aggregation. Their high magnetic content and superior magnetic response kinetics significantly reduce hands-on time compared to conventional isolation methods.

mRNA capture is mediated by sequence-specific complementary hybridization between surface-bound Oligo(dT) and the 3′ poly(A) tail of eukaryotic mRNA. The beads enable efficient enrichment of intact poly(A)+ RNA either from high-quality total RNA, or directly from crude lysates of animal/plant tissues and cultured cells—eliminating the need for prior rRNA/tRNA removal and mitigating the mRNA degradation risk inherent in conventional TRIzol-based extraction workflows.

Features and applications

Features
  • Optimized Oligo(dT) Grafting Density: Balances high capture efficiency with low off-target binding, minimizing non-specific enrichment of rRNA, tRNA, or fragmented RNA to ensure high-purity poly(A)+ RNA recovery.
  • Consistent Physical Performance: Uniform ~1 μm particle size and aggregation-free dispersion guarantee reproducible capture efficiency across batches, with no clogging in automated liquid handling systems.
  • Rapid Workflow Efficiency: High magnetic content enables fast magnetic separation (≤2 minutes per wash step), reducing total hands-on time by >50% compared to column-based mRNA isolation methods.
  • Flexible Input Compatibility: Supports two streamlined workflows: direct capture from crude tissue/cell lysates (eliminating RNA extraction steps) or purification from pre-isolated total RNA, adapting to diverse experimental needs.
  • Intact mRNA Preservation: Avoids harsh organic solvents and multi-step purification inherent in TRIzol workflows, preserving mRNA integrity for downstream full-length transcript analysis.
  • Ecosystem Compatibility: Recovered mRNA is directly compatible with the full AcuGenix™ portfolio of reverse transcription and NGS library preparation kits, enabling end-to-end workflow integration.
Applications
  • mRNA isolation for RNA-seq (bulk, single-cell, and spatial transcriptomics workflows)
  • Full-length cDNA library construction
  • Gene expression profiling (RT-qPCR, digital PCR)
  • Northern blot analysis and primer extension assays
  • Transcriptome studies of low-input or degradation-sensitive samples (e.g., FFPE-derived RNA, circulating tumor cells)
  • Targeted mRNA enrichment for custom NGS panels

Performance data

Comparison of rRNA Depletion Efficiency

BR3R101 AcuGenix™ mRNA Capture Beads: Figure 1 Competitive benchmarking of rRNA depletion performance
Figure 1 Competitive benchmarking of rRNA depletion performance. BR3R101 (AcuGenix™ mRNA Capture Beads) achieved a lower residual rRNA rate than competitor products at both 10 ng and 1000 ng total RNA input levels.

Note: BR3R101 is the catalog number for this product. rRNA accounts for 80–90% of total eukaryotic RNA; low residual rRNA rates directly improve the proportion of on-target mRNA reads, reducing unnecessary sequencing costs.

rRNA Depletion Performance across Different Input Ranges

BR3R101 AcuGenix™ mRNA Capture Beads: Figure 2 rRNA depletion performance across gradient RNA inputs
Figure 2 rRNA depletion performance across gradient RNA inputs. BR3R101 maintained consistently low residual rRNA rates across a broad input range of 10–500 ng total RNA, with no performance degradation.

Note: This input range supports low-input workflows including single-cell sequencing and clinical specimen analysis (e.g., circulating tumor cells, fine-needle aspirates) without requiring protocol re-optimization.

Cross-Species rRNA Depletion Compatibility

BR3R101 AcuGenix™ mRNA Capture Beads: Figure 3 Evaluation of rRNA depletion performance across species
Figure 3 Evaluation of rRNA depletion performance across species. BR3R101 delivered consistent, high-performance rRNA depletion for samples from animals, humans, and plants.

Note: Validated compatibility across common eukaryotic model organisms, clinical samples, and agricultural crop species eliminates the need for species-specific optimization, streamlining multi-species research workflows.

Compatibility with Degraded RNA Templates

BR3R101 AcuGenix™ mRNA Capture Beads: Figure 4 Compatibility test with RNA templates of varying degradation levels
Figure 4 Compatibility test with RNA templates of varying degradation levels. 300 ng of cotton RNA was heat-treated at 72°C for 0–60 minutes to simulate graded RNA degradation, followed by rRNA depletion and library construction. BR3R101 maintained effective rRNA removal even under moderate degradation conditions.

Note: Heat treatment at 72°C is a standard in vitro method for simulating RNA degradation. This performance enables library construction from FFPE-derived RNA, long-term stored samples, or partially degraded specimens, minimizing precious sample waste.
