Product overview

The AcuGenix™ Universal DNA Library Prep Kit is a dual-indexed library preparation solution compatible with all Illumina and MGI high-throughput sequencing platforms. It integrates three critical pre-ligation processing steps—controlled DNA fragmentation, end repair, and 3′-deoxyadenosine (dA) tailing—into a single-tube reaction, eliminating the need for intermediate purification between steps. Users can proceed directly to adapter ligation and library enrichment post-reaction, streamlining the workflow to under 1 hour of hands-on time while minimizing nucleic acid loss for precious low-input samples.

Features and applications

Features
  • True Dual-Platform Compatibility: Validated for direct cluster generation and sequencing on all Illumina and MGI instruments, with no buffer re-optimization required for most applications—eliminating the need for platform-specific library preparation workflows.
  • Streamlined Single-Tube Workflow: Consolidates three enzymatic steps into one closed-tube reaction, removing three rounds of intermediate purification to reduce sample loss for low-input specimens (e.g., FFPE DNA, cfDNA) and minimize aerosol contamination risks from open-tube handling.
  • Broad Input & Sample Compatibility: Supports a wide input range of 100 pg–1 μg total DNA and is compatible with diverse sample types including animal/plant genomic DNA, microbial genomic DNA, FFPE-derived DNA, cell-free DNA (cfDNA), and chromatin immunoprecipitation DNA (ChIP-DNA).
  • Tunable Insert Size: Insert fragment length can be precisely adjusted by varying fragmentation reaction time. The default 150–350 bp uniform size distribution is optimized for paired-end sequencing on both platforms, ensuring balanced coverage for downstream analysis.
  • Ultra-High Purity & Batch Consistency: Each batch is rigorously validated to be free of detectable RNase, DNase, and extraneous nucleic acid contamination, with consistent fragment size distribution across all production lots to guarantee reproducible sequencing performance.
Applications
  • Whole-genome sequencing (WGS) and whole-exome sequencing (WES)
  • Cell-free DNA sequencing (cfDNA-seq) for oncology and liquid biopsy research
  • FFPE-derived DNA sequencing for retrospective clinical studies
  • Chromatin immunoprecipitation sequencing (ChIP-seq) for epigenomic profiling
  • Microbial genome sequencing and metagenomic library preparation

Multi-Species Compatibility

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 1. Multi-species compatibility validation
Figure 1. Multi-species compatibility validation. Six DNA templates from distinct biological sources were processed using BR3D201 (AcuGenix™ Universal DNA Library Prep Kit) for library construction: E. coli (Lane 1), T. thermophilus (Lane 2), phi29 amplification product (Lane 3), λDNA (Lane 4), 10-kb high-fidelity amplicon (Lane 5), and 293T genomic DNA (gDNA, Lane 6). All lanes exhibited comparable band intensity and uniform recovery with no template-dependent variation, confirming that BR3D201 delivers stable, background-independent end modification for heterogeneous and complex genomic samples.

Note: BR3D201 is the catalog number for this kit. Multi-species compatibility is a critical metric for evaluating library prep reagent versatility, supporting workflows with microbial, animal/plant, and clinical sample types.

EDTA Tolerance

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 2. EDTA tolerance of the BR3D201 kit
Figure 2. EDTA tolerance of the BR3D201 kit. Reactions were supplemented with 0–0.8 mM EDTA (Lane 1: 0 mM; 2: 0.1 mM; 3: 0.2 mM; 4: 0.4 mM; 5: 0.8 mM). BR3D201 maintained consistent library size distributions at EDTA concentrations up to 0.2 mM, corresponding to 10 μL of TE-eluted DNA in a 50 μL reaction volume, demonstrating excellent buffer compatibility.

Note: EDTA tolerance prevents enzyme inhibition from residual storage/elution buffers, improving library construction success rates for low-input samples.

Enhancer Mitigates Organic Contaminant Inhibition

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 3. Frag Enhancer mitigates organic contaminant inhibition of BR3D201 fragmentation
Figure 3. Frag Enhancer mitigates organic contaminant inhibition of BR3D201 fragmentation. Reactions were spiked with 0–4 μL Frag Enhancer and low-purity DNA (A260/A230 <1.8; contaminated with guanidine, urea, and phenol from extraction kits). No additive (Lane 1) resulted in complete fragmentation failure; 2, 3, and 4 μL (Lanes 3–5) restored uniform target smear profiles, eliminating the need for pre-purification of inhibitor-containing samples. Frag Enhancer is not included in the BR3D201 library prep kit and is available upon request as a standalone reagent.

Note: Clinical samples (e.g., FFPE, sputum) and environmental samples often contain extraction inhibitors; Frag Enhancer significantly improves library construction robustness for these challenging sample types.

Reduced Amplification Cycles with Single-Step Workflow

DNA InputAmplification Cycles (Total library yield: 1000 ng)
1000 ng1-3
500 ng3-5
100 ng5-7
10 ng9-11
1 ng11-13
100 pg13-15
Figure 4. Fewer amplification cycles required with the BR3D201 kit. BR3D201 was run in its integrated fragmentation/end repair/dA-tailing format across input masses ranging from 100 pg to 1000 ng. Cycle numbers were adjusted to achieve a target library yield of 1000 ng (see accompanying table). BR3D201 consistently required fewer amplification cycles, reducing duplication rates and technical noise while maintaining target yield.

Note: Reduced cycle counts minimize amplification bias and duplicate read fractions, improving the usable data yield of sequencing runs.

Time-Dependent Size Control

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 5. Time-dependent fragment size control with BR3D201
Figure 5. Time-dependent fragment size control with BR3D201. 293T gDNA (0.1, 1, 10, 1000 ng; Lanes 1–4) was fragmented for 7 minutes within the BR3D201 single-step workflow. Electrophoresis showed matched dominant fragment sizes across all input masses, confirming that fragment length is dictated by incubation time rather than DNA input. This enables predictable insert size tuning for any starting mass.

Note: Time-dependent size control is a key advantage of enzymatic fragmentation, simplifying protocol optimization across diverse input amounts.

Library & Capture Parity with Supplier V

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 6. Pan-cancer library and capture parity with Supplier V
Figure 6. Pan-cancer library and capture parity with Supplier V. Lanes: 1 = BR3D201, 1 ng pan-cancer reference material; 2 = BR3D201, 30 ng pan-cancer reference material; 3 = Supplier V, 1 ng; 4 = Supplier V, 30 ng; 5 = BR3N601 hybridization capture reagent + capture workflow. All pre-capture products exhibited matched main peak distributions; Lane 5 retained the profile post-capture. These results confirm BR3D201 is compatible with downstream pan-cancer probe panels for oncology research applications.

Note: BR3N601 is the catalog number for the AcuGenix™ Hybridization Capture Kit. Parity validation confirms BR3D201 can directly replace Supplier V for pan-cancer panel library preparation without workflow re-optimization.

Matched Library Profiles with Superior Pan-cancer Variant Detection

BR3D201 AcuGenix™ Universal DNA Library Prep Kit: Figure 7. Matched library profiles with superior pan-cancer variant detection vs. Supplier V
Figure 7. Matched library profiles with superior pan-cancer variant detection vs. Supplier V. BR3D201 and Supplier V were processed through an end-to-end workflow using 1 ng or 30 ng of pan-cancer reference DNA. Pre-capture size profiles were concordant between both reagents; sequencing analysis revealed BR3D201 had reduced off-target false positives and improved detection of known SNPs/InDels, meeting performance requirements for pan-cancer panel research.

Note: Low off-target rates and high variant detection sensitivity are critical performance metrics for oncology panels. BR3D201 delivers more reliable variant calling results for cancer research workflows. For Research Use Only.
