Product overview

The AcuGenix™ Plant Tissue Direct Hifi PCR Kit is a specialized direct-PCR solution optimized for high-fidelity amplification directly from unextracted plant tissues. It delivers broad compatibility across diverse plant taxa and excellent run-to-run reproducibility. The kit utilizes a proprietary lysis buffer system that rapidly disrupts a wide range of plant materials—including leaf, stem, root, seed, and callus-to release intact genomic DNA within minutes. No prior removal of proteins, RNA, or secondary metabolites (e.g., polysaccharides, polyphenols) is required: the crude lysate is directly compatible with PCR amplification.

Features and applications

Features
  • Streamlined Extraction-Free Workflow: Eliminates time-consuming nucleic acid extraction steps, reducing hands-on time by >70% compared to conventional workflows. The simplified process minimizes aerosol contamination risks from open-tube handling and prevents template loss associated with purification steps.
  • Ultra-High Fidelity Amplification: The 2× master mix contains an engineered antibody-blocked hot-start DNA polymerase with ~154-fold higher fidelity than wild-type Taq DNA polymerase, validated via standardized mismatch rate assays. The enzyme also exhibits exceptional DNA binding affinity and rapid processivity, with robust compatibility for templates containing complex secondary structures or partial degradation commonly found in recalcitrant plant tissues—drastically reducing sequence errors for downstream cloning, sequencing, and genome editing validation.
  • Robust Tolerance to Plant-Derived Inhibitors: The optimized master mix formulation is supplemented with proprietary extension enhancers and specificity promoters, effectively mitigating inhibition from polysaccharides, polyphenols, and other secondary metabolites. It delivers reliable amplification from challenging sample types that typically fail with conventional direct PCR kits.
  • Simplified, Reproducible Operation: All core components (hot-start high-fidelity DNA polymerase, dNTPs, optimized reaction buffer) are pre-mixed in the 2× format. Users only need to add primers and template to initiate amplification, reducing pipetting errors and improving inter-assay reproducibility.
Applications
  • Crop genotyping (SNP, SSR, and InDel analysis)
  • Transgenic event detection in genetically modified (GM) crops
  • Molecular marker development (CAPS, dCAPS, and SCAR marker validation)
  • Germplasm resource identification and diversity analysis
  • Validation of plant genome editing outcomes (CRISPR-Cas9 knock-in/knock-out verification)
  • Template preparation for Sanger sequencing or targeted NGS of plant targets

Performance data

Broad sample compatibility

BR1A503 AcuGenix™ Plant Tissue Direct Hifi PCR Kit: Figure 1 Direct PCR performance across diverse sample types
Figure 1 Direct PCR performance across diverse sample types. BR1A503 (AcuGenix™ Plant Tissue Direct Hifi PCR Kit) enables rapid, extraction-free direct PCR from plants, fungi, high-polysaccharide/high-polyphenol plant species, bacteria, and whole blood samples, eliminating the need for prior nucleic acid purification.

Note: High polysaccharide (e.g., apple, tea) and high polyphenol (e.g., cotton, grape) matrices are common PCR inhibitors in plant workflows. This compatibility addresses amplification failures with recalcitrant plant samples, supporting crop breeding, pathogen detection, and other research applications.

Robust Tissue Lysis Capacity

BR1A503 AcuGenix™ Plant Tissue Direct Hifi PCR Kit: Figure 2 Amplification of multiple tissue types from the same plant species
Figure 2 Amplification of multiple tissue types from the same plant species. BR1A503 efficiently lyses and amplifies diverse tissues from a single plant (including juvenile leaf, mature leaf, stem, root, and seed) without requiring tissue-specific lysis optimization, drastically simplifying workflow setup.

Note: No complex pre-treatment (e.g., grinding, freeze-thaw cycles) is required—simply add the tissue section to the lysis buffer to release intact genomic DNA, minimizing template loss.

Rapid Amplification Kinetics

BR1A503 AcuGenix™ Plant Tissue Direct Hifi PCR Kit: Figure 3 Extension rate evaluation, 2 sec/kb
Figure 3 Extension rate evaluation. BR1A503 achieves an extension rate of 2 sec/kb. When amplifying target genes ranging from 300 bp to 2000 bp in Arabidopsis thaliana (a model plant species), all targets were efficiently amplified, covering the mainstream amplicon size range for plant genotyping and molecular marker development.

Note: This represents a 2–5× improvement over the 5–10 sec/kb extension rates of conventional direct PCR kits, significantly reducing cycling time for high-throughput crop screening workflows.

High-Fidelity Amplification

BR1A503 AcuGenix™ Plant Tissue Direct Hifi PCR Kit: Figure 4 Fidelity benchmarking against competitor products
Figure 4 Fidelity benchmarking against competitor products. The Hifi Master Mix included in BR1A503 delivered superior amplification fidelity compared to all tested commercial competitor products, consistent with the kit’s validated ~154-fold higher fidelity than wild-type Taq DNA polymerase. This minimizes sequence errors for downstream sequencing, cloning, and genome editing validation workflows.

Note: High fidelity is critical for applications requiring accurate amplification, including CRISPR editing site verification and SNP genotyping.
