1 Complete workflow of CircRNA Synthesis Kit
Description: To demonstrate the complete workflow of the Biori CircRNA Synthesis Kit, from DNA template to purified circular RNA. The kit provides all necessary components, including NTPs, transcription buffer, and enzyme mix for in vitro transcription, as well as circularization buffer and RNase R for linear RNA removal via Group I intron self-splicing. The products at each stage—preRNA (linear transcription product), circRNA (after circularization), and circRNA after RNase R digestion—were analyzed by agarose gel electrophoresis to confirm successful circularization and linear RNA removal.

Conclusion: The Biori CircRNA Synthesis Kit enables an integrated and efficient workflow from transcription through circularization and purification, as demonstrated by the successful generation of circRNA via Group I intron self-splicing and effective removal of linear precursors by RNase R digestion, yielding highly enriched circular RNA.
2 circRNA Expression in 293T Cells
Description: To evaluate the expression durability of circRNA prepared using the Biori CircRNA Synthesis Kit, 293T cells were transfected with purified circRNA. eGFP fluorescence was monitored at 24, 48, 72, and 168 h post-transfection to assess the duration and stability of transgene expression.

Conclusion: circRNA prepared using the Biori CircRNA Synthesis Kit mediated high-level eGFP expression in 293T cells at 24, 48, and 72 h post-transfection, as shown in Figure 2. Notably, clear fluorescence was still detectable at 168 h post-transfection, indicating sustained translational activity of the synthesized circRNA beyond the time points shown.
