mRNA Co-transcription and Synthesis Kit, BP-07, supports co-transcriptional synthesis of capped mRNA from T7-promoter DNA templates. The workflow uses T7 RNA polymerase, NTP substrates, and cap analogs to generate 5'-capped RNA according to the instruction manual.

1 mRNA Co-transcription: Template Compatibility and Substrate Flexibility

Description: To evaluate the transcription performance of the Biori mRNA Co-transcription and Synthesis Kit using four DNA templates of varying sizes. Transcription reactions were performed with each template under both natural (UTP) and modified (N1-Me-pUTP) nucleotide conditions. The resulting RNA products were analyzed for yield and integrity.

2 Capping efficiency analysis of Cap1-mRNA prepared with the mRNA Co-transcription and Synthesis Kit

Description: Cap1-mRNA synthesized using the Biori mRNA Co-transcription and Synthesis Kit was processed with the mRNA Capping Efficiency Detection Pretreatment Kit (Cat: BP-05-10) and subsequently analyzed by LC-MS to determine the capping efficiency.

3 eGFP expression of Cap1-mRNA in 293T cells

Description: Cap1-mRNA prepared using the Biori mRNA Co-transcription and Synthesis Kit was transfected into 293T cells to evaluate its translational expression efficiency. eGFP fluorescence was monitored to assess the expression level.
